fa بررسی اثر ضدمیکروبی عصاره اتانلی گیاه Artemisia Turcomanica بر سویه-های مقاوم به آنتی‌بیوتیک کلبسیلا پنومونیه و تاثیر آن در بیان ژن‌های مقاوم به داروی fimH و rmpA Study on antimicrobial effect of Artemisia Turcomanica ethanolic extract into Klebsiella pneumoniae antibiotic-resistant strains and Its effect on rmpA and fimH drug-resistance genes expression تخصصي Special پژوهشي Research زمینه و هدف: با افزایش مقاومت آنتی‌بیوتیکی و گسترش سویه های مقاوم باکتریایی، استفاده از عصاره‌های گیاهی برای درمان، گزارش شده است. هدف از این تحقیق، بررسی اثر ضد باکتریایی گیاه درمنه ترکمنی (Artemisia turcomanica) بر سویه‌های مقاوم  کلبسیلا پنومونیه و تاثیر ان بر بیان ژن‌های مقاوم به داروی fimH و rmpA  بود. روش ها: از 100 نمونه بالینی جمع آوری شده با تست‌های بیوشیمیایی سویه‌های کلبسیلا پنومونیه جداسازی شدند و مقاومت آنتی‌بیوتیک آن‌ها با تست دیسک دیفیوژن بررسی شد. حضور ژن‌های rmpA و fimH  با روش PCR مشخص شد. پس از عصاره گیری از گیاه درمنه ترکمنی، میزان اثر ضدباکتریایی آن در غلظت‌های مختلف بر سویه‌های مقاوم اندازه گیری شد. بیان ژن‌های rmpA و fimH  با روش Real-Time PCR قبل و بعد از تیمار با عصاره گیاهی بررسی شد. داده ها با استفاده از نرم افزار SPSS آنالیز شدند. نتایج: از تعداد  100 ایزوله‌ ، 30 ایزوله  کلبسیلا پنومونیه شناسایی شد و از این 30 سویه، 10 ایزوله مقاوم به چند آنتی‌بیوتیک جداسازی شدند.  ژن های fimH و rmpA بترتیب در هشت و هفت سویه مقاوم، حضور داشتند. مقاوم‌ترین سویه‌ها در برابر عصاره گیاه درمنه ترکمنی، MIC برابر 200 میکروگرم بر میلی‌لیتر داشتند. نتایج Real-Time PCR نشانگر کاهش معنادار در بیان ژن های fimH  و rmpA  بعد از تیمار با عصاره گیاهی بود(P<0.001) . نتیجه گیری: بر اساس این نتایج، درمنه ترکمنی می تواند اثر ضد باکتریایی با کاهش بیان ژن های مقاوم به دارو در باکتری کلبسیلا پنومونیه داشته باشد. برای اثبات نتایج این تحقیق، مطالعات بیشتر نیاز است. Background & Aim: By antibiotic resistance growing and improvement of resistant bacteria strains, was reported useing of plant extracts considering. The aim of this study was to identification the antibacterial effect of Artemisia turcomanica on Klebsiella pneumoniae resistant strains and its effect of fimH and rmpA drug resistance genes expression. Methods: From 100 clinical samples were collected, Klebsiella pneumoniae strains were detected by biochemical tests and their antibiotic resistance was identified by disk diffusion test. The presence of fimH and rmpA genes was found using PCR technique. After of Artemisia turcomanica extraction, its antibacterial effect was evaluated of different concentrations against resistance strains. Expression of fimH and rmpA genes with the Real-Time PCR method, before and after of treatment with herbal extract was investigated. Data by SPSS software was analyzed. Results: Among the 100 isolates, 30 isolates were identified as Klebsiella pneumoniae and from these 30 isolates, 10 isolates with resistance to several antibiotics were selected. The fimH and rmpA genes in eight and seven resistant strains, respectively, were existed. The most resistant strains against the Artemisia turcomanica extract were found with MIC equal to 200 μg/ml. The Real-Time PCR results shown a significant decrease in the fimH and rmpA genes expression after of herbal extract treatment (P<0.001). Conclusion: According of these results, Turkmen herb could be given antibacterial effect with decrease of resistance genes expression in Klebsiella pneumoniae.
