fa بررسی مولکولی گونه های بیماریزای آسپرژیلوس جدا شده زخم پای افراد دیابتی با استفاده از روش Nested PCR Molecular Survey of Pathogen Species of Aspergillus Isolated from Diabetic Foot Lesion Using Nested PCR Method تخصصي Special پژوهشي Research زمینه و هدف: روش PCR حساسیت و ویژگی بالایی را جهت تعیین DNA قارچی و شناسایی سریع اغلب گونه های شایع کلینیکی آسپرژیلوس را ارائه میدهد. هدف از این مطالعه مقایسه کشت با روش nestedPCR برای تعیین آسپرژیلوزیس از زخم پای بیماران دیابتی مراجعه کننده به بیمارستان امام خمینی تهران بود.<o:p /> روش: 65 نمونه زخم های مشکوک به آسپرزیلوزیس موجود در زخم پای دیابتی تحت شرایط کشت و PCR قرار گرفتند. ایزوله های آسپرژیلوس در حد جنس بر روی محیطSGA شناسایی شدند. سپس استخراج DNA انجام و واکنشهای PCR و Nested PCRانجام شد. از بیماران نمونه سرم تهیه و واکنش الیزا جهت یافتن آنتی بادی های ضد جنس و گونه انجام شد. نتایج با استفاده از آزمون آماری آنالیز واریانس تجزیه و تحلیل شدند و P<0.05 معنی دار در نظر گرفته شد.<o:p /> یافته ها: از مجموع 65 نمونه ؛ 39 نمونه بعنوان آسپرژیلوزیس حاصله از روش کشت تشخیص داده شدند. در روشPCR وNested PCR با استفاده از پرایمرهای اختصاصی 41 مورد بعنوان آسپرژیلوس تشخیص داده شدند که از نظر در تشخیص گونه با هم یکسان ولی در تشخیص جنس با هم متفاوت بودند. نتایج الیزا نشان داد که تعداد 39 نمونه مثبت بودند.<o:p /> نتیجه گیری: اگرچه PCR حساسیت کمتری نسبت به سنجش Nested PCR دارد ولی اکثرا شایعترین گونه های بیماریزا یعنی آسپرژیلوس فومیگاتوس را تعیین میکند. نتایج ما نشان میدهند که تفاوت معنی داری مابین نتایج کشت و نتایج nPCRدر شناسایی جنس آسپرژیلوس وجود دارند. Background and Aim: PCR has high sensitivity and specificity for the determination offungal DNA. It is also useful for rapid identification of the most common species of Aspergillus. The purpose of this study was to compare culture method with nested PCR method to determine Aspergillesion in diabetic foot patient in Imam Khomeini Hospital, Tehran.<o:p /> Material and Methods: Sixty-five cases of suspicious diabetic foot lesion Aspergillusis were examined under culture and molecular method. Aspergillus isolates were identified to genus level on the SGA. DNA extraction performed and PCR and Nested PCR reactions were done. Serum samples were collected and ELISA for anti bodies against all species were performed. Results were analyzed using ANOVA and P was considered significant at P<0.05.<o:p /> Results: Of the total of 65 samples, 39 were diagnosed as Aspergillos is obtained from cultures. In PCR and Nested PCR using specific primers, 41 samples were diagnosed as Aspergillus.PCR and Nested PCR for species recognition were equal but differences in gender were recognized. ELISA results showed that 39 samples were positive.<o:p />   Conclusion: Although PCR is less sensitive than nested PCR, but we can recognize the most common pathogenic species, Aspergillus fumigatus.
