Background & Aim: Mesenchymal stem cells (MSCs) are a good candidate for regenerative medicine due to their differentiation and self-renewal properties. However, one of the leading problems is oxidative stress in the target tissue and apoptosis of transplanted stem cells before tissue repair. Pre-treatment of stem cells with antioxidants may make them resistant to oxidative stress. This study was conducted to investigate the antioxidant effect of gingerol on oxidative stress-induced apoptosis and cytotoxicity in mesenchymal stem cells derived from human adipose tissue and rat bone marrow.
Methods: In the present research study, the human adipose tissue and rat bone marrow mesenchymal stem cells were incubated 4 and 6 hours for pretreatment with different concentrations of gingerol (50, 100, 200 and 400 mg / ml) ,then treated with 200 μM H2O2 for 2 hours. Cytotoxicity and apoptosis was analyzed by MTS kit and Annexin V-FITC/PI kit, respectively. Statistical analysis was accomplished by ANOVA and Duncan tests using FlowJo and SPSS 18 softwares.
Results: Gingerol treatment with all concentrations increased the dose and time dependent bioavailability of human adipose tissue-derived mesenchymal stem cells It also decreased apoptosis in rat bone marrow mesenchymal stem cells in a dose- and time-dependent manner.
Conclusion: Gingerolincreases the efficiency of mesenchymal stem cells in tissue regeneration by reducing of oxidative stress.
